Saying Goodbye

Joshua Lederberg (1925—2008) was one of the most influential scientists of the 20th century, and my good friend.

On February 2, 2008, my mentor, beloved colleague, and friend, Joshua Lederberg, succumbed to pneumonia after a long illness. The irony is that he was the ultimate herald of humankind's eternal battle with bacteria and their ability to develop resistance to antibiotics. His prescient essay on the theme "Medical science, infectious disease, and the unity of humankind," published originally in JAMA 1 and reprinted in Current Contents, 2 exemplifies his insight and erudition. To say that Josh was a genius is simply inadequate to describe the breadth of his polymathic talents and interests. Indeed, he was unquestionably one of the most influential scientists of the 20th century.

The editor of a well known life science journal recently asked me to comment on Lederberg's biological work. I declined but referred him instead to several of his laboratory colleagues. From a citationist's perspective, however, there is little doubt about his impact since his published work was cited explicitly in more than 10,000 papers. 3 Yet, the true measure of his impact on science and society is minimally reflected in citation analysis. His seminal discovery of bacterial conjugation 4 in 1946 at the age of 21 helped lay the foundation of molecular biology. Six years later, together with Norton Zinder, came the discovery of transduction. 5 Early on, Lederberg recognized the significance of Oswald Avery's work 6 on pneumococcal DNA. Josh was meticulous in his recognition of predecessors. He treated documentation of the scientific literature in the Talmudic tradition in which he was raised — as a sacred obligation. In an address to science editors published in The Scientist, Josh asserted that "the act of publication is an inscription under oath, a testimony." 7

Neither citation analysis nor a comprehensive acknowledgement index would reveal his multifaceted influence in the fields of artificial intelligence, space science, science policy, scientific communication, information retrieval, and the life sciences. Posted on my personal Web site are dozens of essays in which I extolled Josh as the scientist extraordinaire. 8

His original handwritten letter to me in May 1959 9 recognizing the potential value of a citation index, just months after he received the Nobel Prize, is an extraordinary demonstration of his capacity to see what others could not see. That letter ultimately led to the Genetics Citation Index, which in turn led to the Science Citation Index.

His reverence for the literature and the importance of science criticism led him to play an inspiring leadership role on the board of Annual Reviews. Its Web site highlights his 1987 autobiographical account 10 of the paradigm-breaking discovery of genetic recombination in bacteria. It was Josh who recommended that the Institute for Scientific Information, and later The Scientist, join with Annual Reviews Inc. in sponsoring the National Academy of Sciences annual award for scientific reviewing. 11

My personal bond to Josh was reinforced by our mutual friendship with the sociologists Robert K. Merton and Harriet Zuckerman and countless other creative scholars, including Harold Urey, Luca Cavalli-Sforza, Albert Stunkard, Hilary Koprowski and Carl Djerassi. His encyclopedic network of contacts was invaluable in his role as chair of the editorial board of The Scientist. Josh was an early proponent not only of the idea of a newspaper of science but also open access to the literature. He was a consummate communicator with the public — well illustrated by his years as a weekly columnist for the Washington Post.

His talent as a wordsmith was formidable. His command of the English language and his unusual vocabulary were augmented by his abiding interest in etymology and neologisms. 12 The Oxford English Dictionary lists him as the inventor of the terms "plasmid," 13 "euphenics" 14 and "exobiology" 15 among others. One of his favorite words was "forfend" which, in one sense, epitomized his concern for the survival of humankind through preventive measures. Hence, he had a pivotal role in helping to forestall biological warfare.

His extensive advisory roles in governmental, philanthropic, and industrial organizations alone would warrant a place for him in the pantheon of science. I particularly recall his role in the report of the Weinberg Commission on Scientific Information. 16 Together with William T. Golden, he cochaired the Carnegie Commission on Science and Technology. He played an active role in the Asilomar Conference on Recombinant DNA with Paul Berg and Maxine Singer. The Asilomar Group conducted an early and remarkable exercise in scientific self-control, but Lederberg was also the most outspoken supporter of the looming biotechnology revolution.

A definitive biography of Joshua Lederberg does not exist. While the archives of his papers at the National Library of Medicine, 17 Rockefeller University, and elsewhere will greatly inform the interested scholar, it will be a demanding task for a biographer to capture the full scope of his life and encyclopedic interests. To have shared his friendship for more than 48 years was a unique and fulfilling experience. In this brief tribute it is impossible to digest fully my own shared experiences with Josh. It was a very sad and poignant privilege to be one of the small group of family and friends who said goodbye to Josh at his funeral. I dedicate this essay to his wife Marguerite, his children David and Anne, his brothers Seymour and Dov, his assistant Mary Jane, and his many colleagues at Rockefeller, Stanford, and Annual Reviews.

Major contribution to immunology

Dr. Lederberg made so many contributions that some may not have been given the recognition they deserve. I would like to highlight one such case here. Prevailing immunologic theory regarding self-tolerance was for many decades based on Burnet?s 1949 postulate that there exists a window for tolerance induction early in the life of the animal. While this idea still persists to a limited degree today, it was instead Lederberg?s proposal that has stood the test of time. In a work of remarkable clarity, titled Genes and Antibodies (published in 1959; Science 129: 1649-1653), Lederberg postulated that it is the age of the lymphocyte that matters rather than the age of the animal; i.e. the tolerance window occurs early in lymphocyte development. This process is now commonly referred to as central tolerance, and it is unfortunate that Lederberg is rarely credited for this, one of the most important concepts in immunology.

I remember meeting Joshua Lederberg at a so-called "Scientific Evening" held at the Chemical Industry Institute of Toxiciology. The Scientific Evening was an annual show-and-tell. Dr. Lederberg was on the Scientific Board of the Institute and was visiting the labs for the first time. I was looking forward to merely catching a glimpse of this famous scientist. We had scores of posters tacked to the boards, which lined the maze of hallways. I caught sight of Dr. Lederberg as he walked into the Genetic Toxicology Department's hallway. After a few minutes of looking at posters at the head of the hall, he walked through the crowd to my poster, which described the metabolic capabilities of red blood cells to metabolize aniline to chromosome-damaging intermediates. We introduced ourselves, and he asked me to explain what I had found in my research. We spoke for at least 15 minutes. Needless to say, I was thrilled. As an affirmation to Eugene Garfield's article on the power and range of Dr. Lederberg's mind, Dr. Lederberg asked very good questions from start to finish. And he asked the questions with excitement.



J. Lederberg: a humanist-scientist leader in the 20th century

Being an Egyptian-born student of biological sciences and admirer of genetics at an early age, as an undergraduate, Faculty of Agriculture, Alexandria University,Egypt, my first reading assignment was the classic paper by Lederberg and Tatum, Nature 1946, on bacterial recombinant DNA.This paper inspired me not only during my undergraduate and post-graduate studies in Egypt (National Research Center, Dokki, Giza, Cairo, 1958-1960) but also in my post-graduate training in the USA(Louisiana State University, Baton Rouge,La,1960-1962; The University of Arizona,Tucson,AZ, 1962-1965). Lederberg discovery,along with the classical work of Barbara McClintock on the transposable genetic elements in maize,laid the foundations for molecular genetic research and for the development of genetic engineering tecnology. For their ground-breaking discoveries,both Lederberg and McClintock were justly awarded the Nobel Prize for Medicine and Physiology,in 1958 and in 1983, respectively. As a biologist, I express my sincerest tribute for both of them as outstanding and giant scientists in the 20th century. Garfield's tributal article on the extensive scientific achievements of Lederberg and his humanistic nature illustrated how the world of science in general, and medical sciences in particular, have lost a real world scientific leader.
